I am looking for a way to create one time use tokens that are in nature are like the auto enrollment system but the tokens are deleted after single enrolment.
anyone know how to do this?

No one use tokens, but you could use the Authenticated Access feature, and limit the amount of time the access is available. Under the Manage tab, see Properties>Authenticated Access.

So, you need a bunch of independent links that can be sent out to users, each allowing to sign in into a given course once and only once, right?
The main question here is how do you want to have those tokens passed to their prospective users.
If you simply want a page where all the fifty tokens are displayed in sequence so that one can copy&paste them to email/IM, then it could be done quite easily and based on the existing Authenticated Access functionality; please see bounce.php and tools/access.php for that.
